This study investigated the potential use of cervical mucus characteristics and morphological changes occurring at the cervical opening for determining the timing of estrus and ovulation in ewes subjected to two different durations of progestagen treatment, 7 and 12 days. A total of 18 ewes were randomly allocated into two groups of nine animals each. The ewes in the first group received an intravaginal progesterone sponge containing 20 mg of fluorogestone acetate (FGA) for 7 days, whereas those in the second group received the same treatment for 12 days. At the end of the treatment periods, the sponges were removed, and all animals received 400 IU of equine chorionic gonadotropin (eCG) intramuscularly. Twenty-four hours after sponge removal, a teaser ram fitted with a mating-preventing apron was introduced into the flock for 96 hours. The time at which each ewe accepted the ram was considered the onset of estrus (0 h). Following the onset of estrus, cervical mucus characteristics, morphological changes at the cervical opening, and ultrasonographic examinations were performed simultaneously at 0, 12, 18, 22, 26, and 30 h. The cervical opening was evaluated using a speculum, and hyperemia, edema, and relaxation were scored, while the appearance and amount of cervical mucus were recorded. At the same time points, ultrasonographic examinations were performed to monitor dominant follicle development, and the time at which the dominant follicle disappeared was considered the time of ovulation. Dominant follicle development was monitored by ultrasonography, and follicle disappearance was considered the time of ovulation, which was confirmed by corpus luteum presence nine days later. Additionally, corpus luteum presence was assessed nine days later to confirm ovulation. According to Spearman correlation analysis, a strong, positive, and statistically significant correlation was found between mucus appearance and the timing of estrus and ovulation (r = 0.801; P < 0.001). In contrast, no statistically significant correlations were detected between mucus amount and edema (r = −0.257; P = 0.061) or hyperemia (r = −0.062; P = 0.655) scores and the timing of estrus and ovulation. A moderate, positive, and statistically significant correlation was observed between follicle diameter and the timing of estrus and ovulation (r = 0.485; P = 0.010). In conclusion, the strong and significant association between cervical mucus appearance a9d the timing of estrus and ovulation indicates that this parameter may be useful for evaluating the timing of estrus and ovulation in ewes. Similarly, dominant follicle diameter may contribute to the prediction of estrus and ovulation timing. In contrast, mucus amount, hyperemia, and edema scores were considered insufficient on their own for determining the timing of estrus and ovulation.
