Inula viscosa (L.) Aiton bitkisinin fitokimyasal analizi ve biyolojik aktivitelerinin araştırılması

Özet
Bu çalışma, Inula viscosa (L.) Aiton bitkisinin toprak üstü kısımlarının fitokimyasal incelenmesini; bitki ekstrelerindeki toplam fenolik/antosiyanin içerikleri, fenolik bileşenleri, antioksidan ve antimikrobiyal aktivitelerinin değerlendirilmesini amaçlamaktadır. GC-MS yöntemiyle analiz edilen I. viscosa uçucu yağında toplam 74 bileşen tespit edilmiş, en baskın ana bileşenin %10,60 oranı ile bir oksijenli seskiterpen olan (Z)-α-transbergamotol olarak belirlenmiştir. Hazırlanan hekzan (IVH), aseton (IVA), metanol (IVM) ile direkt petrol eteri (DIVPE), direkt aseton (DIVA), direkt metanol (DIVM), dekoksiyon (IV-def) ve infüzyon (IV-inf) ekstrelerinin sekonder metabolit profilleri LC-MS/MS yöntemi ile analiz edilerek toplam 33 bileşen tanımlanmıştır. Tüm ekstrelerde baskın bileşenlerin ramnositrin (%60-86) ve nepetin-7-glikozit (%5-15) olduğu saptanmıştır. Ayrıca, toplam fenolik madde içeriği en zengin ekstrenin IVA (3838,16 mg/kg) olduğu belirlenmiştir. Toplam fenolik madde miktarı Folin-Ciocalteu yöntemiyle, toplam antosiyanin içeriği ise pH-diferansiyel metoduyla belirlenmiştir. Analizler sonucunda, toplam fenolik içeriği en zengin ekstrenin IVM (702,2 ± 0,12 mg GAE/g) olduğu, toplam antosiyanin miktarı en yüksek içeriğin ise IVA ekstresi (0.465± 0.001 siyanidin-3-glukozit eşdeğeri) olduğu saptanmıştır. Ayrıca bitkiden toplam 4 bileşik (inuviskolit, tomentosin, karabron ve pulişalkonoit C) izole edilmiş ve yapıları NMR (1H-NMR, 13C-NMR, COSY, HMQC, HMBC) teknikleri ile aydınlatılmıştır. Ekstrelerin antioksidan kapasiteleri DPPH (2,2-difenil-1-pikrilhidrazil) radikal giderme, bakır (II) iyonu indirgeme antioksidan kapasitesi (CUPRAC) ve demir (III) indirgeyici antioksidan güç tayini (FRAP) yöntemleriyle değerlendirilmiştir. DPPH ve CUPRAC testlerinde en yüksek antioksidan aktiviteyi IVM ekstresi, FRAP testinde ise IV-def ve IVinf örnekleri göstermiştir. Ekstrelerin antioksidan potansiyellerinin toplam fenolik içerikleriğle doğru orantılı olduğu saptanmıştır. Antimikrobiyal çalışmalar; ekstrelerin gram-pozitif bakteri ve fungal türlere karşı etkili, gram-negatif bakterilere karşı ise direnç gösterdiğini kanıtlamıştır. Özellikle orta ve yüksek polariteli ekstrelerin daha yüksek inhibitör aktivite sergilediği görülmüştür. Sonuç olarak, I. viscosa ekstrelerinin zengin fenolik içerikleriyle güçlü antioksidan ve antimikrobiyal potansiyel taşıdığı saptanmıştır.
This study aims to investigate the phytochemical profile of the aerial parts of Inula viscosa, determine total phenolic and anthocyanin contents, analyze phenolic components, and comprehensively evaluate the antioxidant and antimicrobial activities of these extracts. Seventy-four components were identified in the I. viscosa essential oil analyzed by GCMS, with (Z)-α-trans-bergamotol, an oxygenated sesquiterpene, being determined as the most dominant main component at a rate of 10.60%. Secondary metabolite profiles of hexane (IVH), acetone (IVA), methanol (IVM), direct petroleum ether (DIVPE), direct acetone (DIVA), direct methanol (DIVM), decoction (IV-def) and infusion (IV-inf) extracts were analyzed via LC-MS/MS, identifying 33 components. Rhamnocitrin (60- 86%) and nepetin-7-glucoside (5-15%) were the dominant compounds in all extracts. IVA (3838.16 mg/kg) had the highest phenolic content by LC-MS/MS. Spectrophotometric analyses showed the highest total phenolic content in IVM (182.64 ± 0.03 mg GAE/g) and the highest total anthocyanin content in IVA (0.465 ± 0.001 cyanidin-3-glucoside equivalent).Additionally, four compounds (inuviscolide, tomentosin, carabrone, and pulichalconite C) were isolated from the plant, and their structures were elucidated using NMR (1H-NMR, 13C-NMR, COSY, HMQC, HMBC) techniques. Antioxidant capacities were evaluated using DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging, CUPRAC (Cupric Ion Reducing Antioxidant Capacity), and FRAP (Ferric Reducing Antioxidant Power) assays The IVM extract showed the highest antioxidant activity in DPPH and CUPRAC assays, while the IV-dek (decoction) and IV-inf (infusion) samples showed the highest activity in the FRAP assay. Overall, antioxidant potential correlated positively with total phenolic content. Antimicrobial assays proved that the extracts were effective against gram-positive bacteria and fungi, while gram-negative bacteria showed resistance. Extracts with medium to high polarity exhibited superior inhibitory activity. In conclusion, I. viscosa extracts possess significant antioxidant and species-specific antimicrobial potential due to their rich phenolic content.
