Katı substrat fermentasyonu koşullarında aspergillus aculeatus NRRL 358’da ekstraselüler β-glukozidaz üretimi, saflaştırılması ve karakterizasyonu

Özet
Bu çalışmada, arpa samanı kullanılarak hazırlanmış KSF ortamında geliştirilen Aspergillus aculeatus NRRL 358’dan ASÇ ve HEK yöntemleri kullanılarak β-glukozidaz enzimi saflaştırılmıştır. A. aculeatus’un KSF ortamında en yüksek β-glukozidaz aktivitesine sahip olduğu optimum koşullar belirlenmiştir. Nemlendirme sıvısı Sodyum Dihidrojen Fosfat (NaH2PO4); pH’ın 7,0; sıcaklığın 25°C ve inkübasyon süresinin ise 10 gün olduğu saptanmıştır. Elde edilen β-glukozidaz enzimi HEK uygulaması sonucunda %43,08 verim ile 23,85 kat saflaştırılmıştır. β-glukozidazın saflığının ve alt birim sayısının kontrolü SDS-PAGE ve NATIVE-PAGE yöntemleri kullanılarak yapılmıştır. NATIVE-PAGE elektroforezi sonucunda 170 kDa molekül ağırlığına sahip olduğu, SDS-PAGE işlemi sonucunda ise enzimin 70 kDa ve 100 kDa olmak üzere iki alt birimden oluştuğu tespit edilmiştir. Enzimin aktivitesinin tespit edilmesinde pNPG substratı kullanılmış, pH 4,0’de ve 55°C’de en yüksek aktiviteye ulaştığı belirlenmiştir. Saflaştırılmış β-glukozidaz enziminin 45, 55, 65 ve 75°C sıcaklıklarda 120 dk inkübasyona bırakılmasıyla yapılan aktivite ölçümleri sonucunda sırasıyla %99,6; %99,8; %98,6 ve %31,87 oranında aktivite gösterdiği, 85°C sıcaklıkta ise 36 dk boyunca aktivitesini kaybetmediği tespit edilmiştir. Saflaştırılan β-glukozidaz enzimi kinetik parametreleri yönünden de incelenmiş, Km ve Vmax değerleri sırasıyla 2,8571 mM ve 14285,71 mM olarak hesaplanmıştır. Bunlara ek olarak, D(+)-glukoz ve δ-glukonolakton inhibitörlerinin saflaştırılan β-glukozidaz enzimine karşı inhibisyon mekanizmalarının yarışmalı tipte olduğu belirlenmiştir. D(+)-glukozun IC50 değeri 11,49 mM ve Ki değeri 1,7857x10-3 olarak, δ-glukonolakton inhibitörünün IC50 değeri 13,64 mM ve Ki değerleri ise 7,143x10-5 ve 1,07145x10-4 mM olarak belirlenmiştir.
In this study, the β-glucosidase enzyme was purified from Aspergillus aculeatus NRRL 358, which was grown on a KSF medium prepared using barley straw, using ASÇ and HEK methods. The optimal conditions under which A. aculeatus exhibited the highest β-glucosidase activity in the KSF medium were determined. The buffer solution was Sodium Dihydrogen Phosphate (NaH₂PO₄); the pH was 7.0; the temperature was 25°C; and the incubation period was 10 days. The β-glucosidase enzyme obtained was purified 23.85-fold with a yield of 43.08% using the HEK method. The purity and subunit number of the β-glucosidase were verified using the SDS-PAGE and NATIVE-PAGE methods. NATIVE-PAGE electrophoresis revealed a molecular weight of 170 kDa, while SDS-PAGE analysis indicated that the enzyme consists of two subunits of 70 kDa and 100 kDa. The enzyme’s activity was determined using the pNPG substrate, and it was found to reach its highest activity at pH 4.0 and 55°C. Activity measurements conducted by incubating the purified β-glucosidase enzyme at 45, 55, 65, and 75°C for 120 minutes revealed activity levels of 99.6%, 99.8%, 98.6%, and 31.87%, respectively, and it was found that the enzyme retained its activity for 36 minutes at 85°C. The kinetic parameters of the purified β-glucosidase enzyme qwwew examined, and the Km and Vmax values were calculated to be 2.8571 and 14,285.71 mM respectively. In addition, it was determined that the inhibition mechanisms of D(+)-glucose and δ-gluconolactone inhibitors against the purified β-glucosidase enzyme were of the competitive type. The IC50 value of D(+)-glucose was 11.49 mM and the Ki value was 1.7857×10⁻³, while the IC50 value of the δ-gluconolactone inhibitor was 13.64 mM and the Ki values were 7.143×10⁻⁵ and 1.07145×10⁻⁴ mM.
