In vitro effects of some herbicides and fungicides on human erythrocyte carbonic anhydrase activity

Özet

The effects of Imazethapyr, 2,4-D dimethylamine salt, Glyphosate isopropylamine salt and Propamocarb HCl, which are commonly used in agricultural fields, have been investigated on human erythrocyte carbonic anhydrase isoenzymes (hCA-I, hCA-II) in vitro. Isoenzymes employed in the study were purified by using Sepharose-4B-L-tyrosine-sulphanylamide affinity gel and the purity was confirmed by SDS-PAGE. In vitro CA enzyme activity was determined colorimetrically using the CO2 hydration method of Maren. All the pesticides evaluated inhibited the activity of isoenzymes to various degrees. IC50 values of chemicals caused inhibition were determined by means of activity percentage-[I] diagrams. These values of Imazethapyr, 2,4-D dimethylamine salt, Glyphosate isopropylamine salt and Propamocarb HCl, for CA-I and CA-II were 0.093 mM, 0.463 mM, 0.617 mM, 0.46 mM and 0.152 mM, 0.628 mM, 0.904 mM, 0.62 mM, respectively. Imazethapyr was the most effective inhibitors for CA-H isoenzyme.
