dc.description.abstractUp Regulated Gene (URG-4) veya hücre poliferasyonunun upregülatörü (URGCP) olarak da bilinen URG-4, 7. kromozomun (7p13) kısa kolu üzerinde lokalize olan bir onkogendir. İlk olarak hepatosellüler karsinoma hücrelerinde (HCC) saptanmıştır. Yapılan çalışmalarda URG-4’ün overkespresyonunun birçok kanser türünde kötü prognoz etkisi gösterdiği ve farklı tümörlerde aşırı eksprese edilerek tümör gelişiminde rol oynadığı görülmüştür. URG4’ün birçok kanser çeşidinde proliferatif ve metastatik karakterine etkisi gözlemlenmiştir. Bu tez kapsamında URG-4/URGCP geninin prostat kanseri üzerinde metastatik karakterlerine etkisinin belirlenmesi ve literatürdeki eksikliğin giderilmesi amaçlanmaktadır. Bu çalışma ilk olarak PC-3 (İnsan Prostat Kanseri Hücre hattı) hücrelerinin büyütülmesi ile başlamaktadır. Linerize olmuş URG-4/URGCP+pCDNA3.2 vektör ve kontrol grubu olarak boş vektörün PC-3 hücrelerine transfeksiyonu sağlanmıştır. Overekspresyon çalışmaları mRNA ve protein seviyesinde doğrulandıktan sonra, URG4/URGCP genine spesifik ekspresyon primerleri haricinde metastatik marker genler olan EKaderin, Vimentin, Osteopondin ve Twist kullanıldı. Apoptotik marker genler olarak da Bcl2, BAX, BAK ve BAD kullanıldı. Proliferasyonu incelemek amacıyla ise Siklin D1 ve p27 marker genler kullanılarak Real-Time PCR reaksiyonu gerçekleştirildi. Daha sonra EKaderin, Vimentin, Bcl-2 ve BAX antikorları ile protein seviyesinde western blot deneyleri yapıldı. URG-4/URGCP aşırı ifadesinin PC-3 hücrelerindeki hücresel karakterizasyonunu belirlemek amacıyla MTT, yaralanma, koloni formasyon deneyleri gerçekleştirildi. Tüm bu çalışmaların sonucunda prostat kanseri hücrelerinde URG-4/URGCP geninin overekspresyonunun metastaz ile ilişkisinin belirlenmesi amaçlanmaktadır. Çalışmaların sonucunda mRNA ve protein düzeyinde URG-4 overekspresyonunun proliferatif ve metastatik etkisinin; hücresel düzeyde ise invazyon yeteneğinin arttığı belirlenmiştir. Bu tez kapsamında elde edilen verilerin, ilerleyen kanser araştırmalarında, araştırmacılara yol göstereceği öngörülmektedir.
dc.description.abstractURG-4, also known as the Up-Regulated Gene (URG-4) or the Up-Regulator of Cell Proliferation (URGCP), is an oncogene localized on the short arm of chromosome 7 (7p13). It was first identified in hepatocellular carcinoma (HCC) cells. Studies have shown that URG-4 overexpression is associated with poor prognosis in many types of cancer and plays a role in tumor development by being overexpressed in various tumors. The effect of URG4 on the proliferative and metastatic characteristics of many cancer types has been observed. The aim of this thesis is to determine the effect of the URG-4/URGCP gene on the metastatic characteristics of prostate cancer and to address a gap in literature. This study begins with the cultivation of PC-3 (human prostate cancer cell line) cells. The linearized URG4/URGCP+pCDNA3.2 vector and an empty vector (as a control) were transfected into PC3 cells. After overexpression studies were validated at the mRNA and protein levels, in addition to URG-4/URGCP gene-specific expression primers, metastatic marker genes such as E-cadherin, vimentin, osteopontin, and Twist were used. Bcl-2, BAX, BAK, and BAD were also used as apoptotic marker genes. To investigate proliferation, Real-Time PCR reactions were performed using the marker genes Cyclin D1 and p27. Subsequently, Western blot experiments were conducted at the protein level using antibodies against E-cadherin, Vimentin, Bcl-2, and BAX. MTT, wound healing, and colony formation assays were performed to determine the cellular characterization of URG-4/URGCP overexpression in PC-3 cells. The aim of all these studies is to determine the relationship between URG4/URGCP gene overexpression and metastasis in prostate cancer cells. The results of the studies revealed that URG-4 overexpression at the mRNA and protein levels enhances proliferative and metastatic effects, while at the cellular level, it increases invasion capacity. It is anticipated that the data obtained in this thesis will guide researchers in future cancer studies.
