CZUY/CIN’in test edilen bakteri suşlarına karşı güçlü antibakteriyel aktivite gösterdiği belirlenmiştir. CZUY/CIN’in gentamisin ve tetrasiklin ile kombinasyonlarında sinerjik etki gözlenmiştir. Sonuç: CZUY ve ana bileşeni CIN’in güçlü antibakteriyel özellikleri ile topikal antibiyotiklerle gösterdiği sinerjik etkileşimler, doğal ürünlerin giderek büyüyen antibiyotik direnci sorununa çözüm arayışında yeni antibakteriyel seçenekler sunabileceğini göstermektedir
Objective: With the continuous emergence of new bacterial strains resistant to antibiotics, the effectiveness of antibiotics has declined, making antibiotic resistance a global public health concern. Phytochemicals may represent promising adjuvants for antibiotics. The aim of this study is to investigate the synergistic interaction between Cinnamomum zeylanicum essential oil (CZEO) and its major component, cinnamaldehyde (CIN), with topical antibiotics. Methods: C. zeylanicum essential oil was obtained by water distillation and its major component was identified through gas chromatography-mass spectrometry (GC-MS) analysis. The antibacterial activities of the essential oil and CIN were determined using the Kirby-Bauer disk diffusion method and the broth microdilution method. A multidrug-resistant Staphylococcus aureus clinical isolate and the reference strain S. aureus ATCC 29213 were used as Gram-positive bacteria; a multidrug-resistant Klebsiella pneumoniae isolate and Escherichia coli ATCC 25922 were used as Gram-negative bacteria. The synergy between CZEO, CIN, and topical antibiotics was evaluated using the checkerboard and time-kill methods. Results: According to GC-MS analysis, CZEO contained 57.41% cinnamaldehyde (CIN), 31.65% cinnamyl acetate, and 3.75% coumarin. The inhibition zones for CZEO and CIN against all bacterial strains ranged between 20– 35 mm and 19–30 mm, respectively; the MIC values were between 0.125–0.5 mg/mL and 0.125–1 mg/mL, respectively. CZEO and CIN demonstrated strong antibacterial activity against the tested bacterial strains. Synergistic effects were observed in combinations of CZEO/CIN with gentamicin and tetracycline. Conclusion: The strong antibacterial properties of CZEO and CIN, along with their synergistic interactions with topical antibiotics, suggest that natural products may offer new antibacterial options in the fight against the problem of antibiotic resistance.
