Özet

GST is the most important detoxifying enzyme of environmental pollutants, such as carcinogenic matters, heavy metals, some of drugs and pesticides. In this study, hydrophobic gel having a chemical structure of Sepharose 4B-L-tyrosine naphthylamine (STN), for the first time, was used for the purification of Broiler chicken glutathione S-transferase (bcGST). The effects of pesticides namely, Delmetrin (R), Mamba Turbo (R), Alpgor (R) and Petra (R) on purified bcGST activity were determined. Among the pesticides tested, Delmetrin (R) and Petra (R) had very strong inhibitory effects on bcGST with IC50 values of 28 mu M, 45 mu M, respectively, but the activity of enzyme was poorly inhibited by Mamba Turbo (R). On other hand, Alpgor (R) with dimethoate had no inhibitory effect on bcGST at the working concentration (20-60 mu M).
